The copper-free click chemistry, strain-promoted azide-alkyne cycloaddition (SPAAC) is a bioorthogonal reaction that utilizes a pair of reagents, a cyclooctyl and an azide compound, which react efficiently with each other while remaining inert to natural functional groups such as hydroxyl, amino and thiol groups.SPAAC is capable of labeling a wide variety of biomolecules in water and other complex chemical environments by forming stable triazoles without the need for any auxiliary reagents. Among the many known cyclooctyl compounds, dibenzocyclooctyl compounds (DBCO) are a class of reagents with fairly fast kinetics and good aqueous buffering stability. DBCO-succinimide esters are a commonly used reagent to introduce DBCO groups into biomolecules through amino modification.
